86million exp to reach lvl 121 from 120. (Actually this is the total, so you would need 78million exp since lvl 120 is ~ 8million exp)

1) Will I loose red portal near to Anya (5th town) if I kill Nihlatak and don't activate Halls of Pain portal?
2) Experience for 121 lvl can I get only from signets of experience? Where can I get so many signets? Also I heard that I can get exp from Destruction Cows (Wirt's Leg + Tome of Town Portal → Cow Level portal). Is it true? How many cows should I kill to reach 121 lvl. As I understand after Save and Exit I loos all exp if I did't get 121 lvl. Will it be enough to kill all cows in Cow Level before Save and Exit to reach 121 lvl? And only Cow level? I heard about duncraig too.
3) Is there exists any reason to farm Nihlatak on destruction difficulty insted of Nihlatak on terror difficulty? Are they both drop 1 signet of learning (this is my main goal to go there)? How often does Nihlathak drop SU items on destruction difficulty (SU /per 100 runs, my magic find is 450%)? Will I get more SU items if set "players 2" or 4, 8? Does it make sence or better I should go to Tran Athula/another uber for SU?

1. I was wrong, see below.
2. You don't NEED signets of experience, but they can help. Any monster will give you experience. People suggestion DCows because it has the highest rate of experience and is rather easy to farm. And no, you don't lose your experience when you leave the game. It just doesn't show up on the bar.
3. Nihlathak on Destruction drops signets of learning (Terror Nihlathal doesn't) and has a chance to drop SUs. I'm not sure how many drops you'd get. You would get better drops with /players, but if you're on singleplayer, then you can't get to level 121 or above, as level 121 and above is on the realm only.
